Specifications for Isothermal Titration Calorimeter
The system must be complete with control unit, degassing
station, filling & cleaning assemblies, injection syringes, other start-up
accessories, necessary software/s for instrument control, operation analysis of
data, viewing and printing enabling accurate determination of thermodynamic
parameters such as binding constants, reaction stoichimetry, enthalpy, entropy
etc. with the following detailed specifications:
Operating temperature range: 2 o C to
80 o C
Temperature stability
± 0.005 o C measured at
different time periods
Sensitivity: High sensitivity with base line noise
level around 0.5 nCal / sec with a 2 second data filter for ultra sensitive
studies
Minimum Response Time (high gain): 20 seconds
Minimum detectable heat effect 0.1 μJ
Mixing speed and Response: Variable mixing speed
and variable response times
Binding affinity: The unit must have the
capability to directly measure binding constants accurately in the range of 102
to 109 M-1 and tight binding constants by displacement
reactions in the order higher than 109 M-1
Heat compensation
to cells should be via power
feedback only for accurate, sensitive heat determination and rapid
response. No solid state heating and cooling systems.
Cell type: Non-capillary, fixed in place –
non-removable, with low working sample volume of 1.5 ml or lower. They should
be enclosed in an adiabatic chamber
Cell design:
Coin-shape - providing
large surface area for the peltier elements and makes it easier to attach
them. Perfect Cell Geometry (coin shaped cell). Fast Equilibration Time. Fast
Response Time.
Cell Material: should be made up of Hastelloy
material which is very inert. No reaction with S-containing groups or
reducing agents. Should have Excellent Chemical resistance. Should work at pH
range 2 – 12 of solvents.
The System should have three user selectable modes of
operation (Response Times) high gain, low gain and medium gain - allowing the
fastest re-equilibration between injections, there by providing the shortest
experimental times.
There should be no use of external water bath etc. The
device should be internal (peltier type).
The system should have ThermoVac for degassing.
Required Computer to form an integral part of the supply.
The software should be Origin®, capable of providing user
selectable binding models and data merging like: • Single Site • Two
Site • Sequential Site • Competitive Site • Enzyme Kinetics
• Displacement
•
Dissociation.
For multiple run analysis.
Instrument operation manuals (English language) should be
included in the standard scope.
Trained technicians should install the instrument and ensure
it is operating correctly and within specifications. User training at site
should also be provided.
Users lists of similar equipment supplied in India should be
provided (Enclose full list of users in India). Vendor should have couple
of users available in last 5 years in the eastern zone of India.
A detailed technical
compliance is required with supporting data where possible to show compliance
