About the Topic: Polyvalent cationic cosolutes
induce DNA complexation, compaction and protection. Our physico-chemical
research on various mixed systems of DNA and cationic amphiphiles and polyions
in bulk and at interfaces will be outlined.
Interaction of DNA with simple cationic surfactants will
be reviewed, followed by DNA interaction with cationic-anionic mixtures.
These also include interaction with thermodynamically stable catanionic
vesicles, built on classical and aminoacid-based surfactants. Through phase
map determinations and cryo-transmission electron microscopy, an overview
from phase behaviour to microstructure will be presented for several cases.
We will also address DNA compaction and decompaction phenomena
in different mixed systems, and their dependence on the amphiphile hydrophobicity
and mixture.
Modelling of DNA compaction by polycations, by means of
Monte Carlo simulations, was also considered. Results clearly indicate
the crucial role of electrostatic interactions in the compaction process.
DNA adsorbs on a hydrophobic surface. The adsorbed amount
is low and the conformation extended. On addition of a cationic surfactant,
the adsorbed amount increases strongly at the same time as DNA compacts.
As the surfactant is removed, DNA decompaction occurs. On a hydrophilic
anionic surface DNA does not adsorb but adsorption can be induced by a
cationic surfactant.
Various aspects of the interfacial behaviour of mixed
DNA-surfactant systems are presented. These include the study of adsorption
on hydrophobic and hydrophilic surfaces by ellipsometry, and the forces
between DNA-covered surfaces by surface force apparatus and atomic force
microscope. Different surfactant types are studied including the combination
of cationic and anionic surfactant. Both thermodynamic and kinetic aspects
are included.

Water is a unique liquid. A detailed
picture of motion of atoms and hydrogen bond network in water has been
obtained recently using large scale computer simulations. It explains properties
of water in unprecedented details. For instance, freezing of water has
been explained for the first time (Ohmine, Nature 416 (2002) 409). Some
of these results will be presented mostly through movie pictures.

About the Speaker :
Professor Geradus 't Hooft along
with Professor Martinus J.G. Veltman, received the Nobel
Prize in
Physics
in 1999 for setting up a functioning theoretical machinery to perform
calculations with non-Abelian gauge theories.
The two researchers are awarded
the Nobel Prize for having placed particle physics theory on a firmer mathematical
foundation. They have in particular shown how the theory may be used for
precise calculations of physical quantities. Experiments at accelerator
laboratories in Europe and the USA have recently confirmed many of the
calculated results.
Unifying the four basic forces --
gravitational, electromagnetic, weak and strong -- has been the holy grail
of theoretical physics. The electromagnetic force keeps electrons in orbit
and helps the atoms to survive, while the weak force keeps the sun burning
by converting protons into neutrons which are then "baked" into helium
nuclei. The combined elcetroweak interaction developed in the sixties was
difficult to accept because no meaningful calculations could be done with
it. The work of 't Hooft and Veltman in the early seventies changed the
scenario and truly unified these two forces.
The 'holographic principle' developed
by 't Hooft in the mid eighties has become the primary tool in tackling
the final unification process -- that of gravity and quantum mechanics.
These, along with numerous other pioneering works, make Professor 't Hooft
one of the greatest theoretical physicists of all time.
